What Nissl Staining Shows
Nissl staining highlights basophilic structures such as rough endoplasmic reticulum and ribosomes in neuronal somata, making it a quick and reliable way to visualise cell bodies and cytoarchitecture across brain regions.
Common Dyes
Cresyl violet, thionin and toluidine blue are the most common Nissl dyes. They bind nucleic acids and produce crisp, blue-purple staining of neuronal nuclei and cytoplasm.
Basic Protocol
- Mount sections on coated slides and allow to dry and adhere.
- Stain with the Nissl dye for the optimised time (typically 1–10 minutes).
- Differentiate in ethanol to remove excess dye until background is clean.
- Dehydrate, clear and coverslip with a permanent mountant.
Quality Considerations
Uniform thickness and consistent staining times are critical when comparing cell counts or densities between groups. Stain batches together and use the same differentiation conditions to keep signal comparable.
Applications
Nissl staining is widely used for anatomical mapping, volumetric analysis and estimating neuron numbers — often as the structural reference paired with other markers or with brain atlases.
